Stimulatory effect of IL-1 secreted by fibroblasts taken from periodontitis  lesion

Interlukin 1 is an inflammatory factor which has a local and generalized  effect by its secretory

type through the circulation. Fibroblasts, on the other hand , are considered of great

importance in connective tissue metabolism including periodontal tissues . They secret

interlukin 1  with other additional substances .

The purpose of our study was to isolate these fibroblasts taken from periodontitis location and

from normal periodontal tissues and grow them in vitro. Then to compare the secretion of

Interlukin 1 from fibroblasts obtained from the normal source and the periodontal disease

source .If difference is found,  this will support the theory of the presence of subpopulation of

fibroblasts that have different functional parameters that lead to changes in the tissue matrix

which clinically manifests as periodontitis.

Fibroblast are isolated from normal and abnormal  periodontal tissues and then grown in

suitable media , then The fibroblast metabolites are harvested and added to the thymocytes

which are incubated with  radio active thymidine.

Results showed that  radioactive Thymidin taking cells count after two days of incubation with

the metabolites from periodontitis source was 2920 +/- 185 and after four days it became 5500

+/- 227 , while the count  after two days of incubation with the metabolites from fibroblast

obtained from normal periodontal tissues was 886+/-142and 1239 +/- 133 after four days of

incubation respectively . The difference is considered in accordance with the student T test a

significant one . This  supports the suggestion that gingival connective tissue pathogenesis

involves not cellular injury but selection , by unique conditions existing within the affected

tissue, of unusual fibroblast subpopulation characterized by functional parameters that lead to

changes in the tissue matrix .
